I can only speak from my experience

Although I have found most speakers to come around substantially after a few hours there have been major exceptions. Speakers that I thought were fairly bad after the first five to ten hours were left on at moderately high volumes with no one around for an additional sixty hours. When I came back the speakers that had sounded worse than mediocre were sounding great. Since I wasn't there listening to the process there was no acclimatizing to the sound. At lower volumes I can see how it could take substantially longer. Roy Johnson from Green Mountain Audio encourages this sort of relatively high volume break in.
Hundreds of hours is outside my experience but that is not to say that I don't believe it could happen.
