Distortion.
Harmonic distortion, or basically differences in the harmonics produced. Also, differences in frequency response.
Plugging in a 6550 into a socket that had KT88's and not changing anything might be sort of overdoing it for a 6550.
One should also consider the reflected impedance of the output transformer when switching tube types around. This will also make a difference in the sound. Using a transformer designed for a specific tube will make a huge difference.
The main reason people only change tubes to change the "voice" of their amplifier is because it's easy and requires little thought. That is, until someone plugs a 5U4 into an output tube socket.
It might be the next step in changing the sound of an amplifier to consider different power supply designs, different output transformers, different coupling caps, etc.
There's a whole world of modifications that can be done. It's just that swapping out tubes is easy and quick.
