Hi ned,you could do either to measure volume. If you leave them in you'll need to add a bit to the volume measured to allow for the volume between the driver frame and cone. If you take the drivers out you'll need to subtract the volume of the drivers.
The correct size port can be calculated using the carstereo site pages. a bigger port is better, and also longer, but you need to subtract the volume of the port from the box volume. Always compromises.
OR put the port on the outside (which would be way pimped). see link for a fellow aussie bass head.
don't forget to line the inside with fake fur.
