Whether the tubes will make any difference at all to the gain depends on the circuit configuration. You haven't said what the amp is - that would help the Asylum to help you!Most preamps and amps use a voltage divider in the feedback loop to set the signal gain, so changing the tubes will not change the gain. You would have to change the appropriate resistor, although in some cases this can reduce the stability of the circuit and cause it to ring or oscillate.
On the other hand, if the gain of circuit depends only on the tube parameters there is space for manoeuvre, but then you have the problem of having to match the tubes for the two channels closely.
