Transconductance is as far as I know the best parameter by which to match tubes. Transconductance (Gm) is defined as the change in output current for a given change in input voltage (with plate voltage held constant). Your tester should surely be able to give very accurate readings of Gm based on the actual operating conditions of your amplifier. I don't think that measuring current at only one operating point is necessarily proof that two tubes are or are not matched, unless the differences are huge.
